Using holoenzyme of recombinant human arginase I, biosensoric and enzymatic approaches for assaying L-arginine (Arg) have been developed. Biosensor is based on using two enzymes (arginase and commercial urease) and amperometric detection of ammonium ions monitored by chemosensing Polyaniline-Nafion membrane. Enzymatic approaches are based on fluorimetric detection of urea (monoenzymatic approach) or ammonia (bienzymatic approach). Apoenzyme of Mn2+- dependent arginase has been used for very sensitive analysis of manganese(II) and cobalt(II) ions after reconstruction of active holoenzyme at the presence of analyzed ions at different pH values.
